HP zorbax HOLC columns b34 User's Guide and Manual
Summary
This chromatography system is specifically designed for the highly sensitive analysis and metabolic profiling of polar compounds, such as nucleotides from DNA and RNA hydrolysates. It allows researchers in biochemistry and medicine to efficiently analyze base pair composition, helping investigate both normal metabolic states and potential disease abnormalities. The stable column maintains strong performance even under alkaline conditions utilizing ion-pairing chromatography techniques essential for these delicate molecules.

A Rapid Communication of Applications and Techniques
Analysis of nucleic acids in biochemistry and medicine grew as a need for rapid determination of base pair composition of DNA and RNA hydrolysates. In medicinal chemistry, nucleotide analysis is of great interest for investigating metabolic states in both the normal metabolism of nucleic acids and abnormalities caused by disease.

ZORBAX® Eclipse XDB-C8 (3.5 µm, 4.6 x 75 mm) (P/N: 966967-906) Mobile Phase: 93% 50 m M Sodium Phosphate (p H 7.0 with Phosphoric Acid) / 10 m M Tetrabutyl ammonium bromide : 7% Acetonitrile Injection volume 1 µL, 1.0 m L/min, 35°C, Detect. UV (254 nm)
HIGHLIGHTS Good efficiency and peak shape can be obtained for very polar compounds using ion-pairing chromatography at elevated p H with Zorbax Eclipse XDB-C8. Nucleotides, due to degradation at low p H and must be analyzed at high p H. Zorbax Eclipse XDB-C8 is very stable with mobile phases even at an elevated p H of 7.0, due to double-endcapping, which protects the silica surface.

Frequently Asked Questions
What column is recommended for nucleotide analysis?
ZORBAX Eclipse XDB-C8 (3.5 μm, 4.6 x 75 mm, P/N: 966967-906).
Why should pH be maintained at 7.0 for nucleotide analysis?
Nucleotides degrade at low pH and must be analyzed at high pH.
What mobile phase is used for nucleotide separation?
93% 50 mM Sodium Phosphate (pH 7.0) / 10 mM Tetrabutyl ammonium bromide : 7% Acetonitrile.
How is the Zorbax Eclipse XDB-C8 column stabilized at high pH?
Double-endcapping protects the silica surface, ensuring stability at pH 7.0.
